白木通组织培养及快速繁殖

摘 要:

以白木通的带芽茎段为外植体, 研究植物生长调节剂对其离体培养及植株再生的影响, 建立了木通离体快繁技术体系。结果表明: 带芽茎段的较佳消毒灭菌方法为75%的酒精浸泡30 s, 再用2%的次氯酸钠溶液浸泡15 min; 腋芽诱导的最适宜培养基为WPM+1.0 mg•L-1 6-BA+0.5 mg•L-1 IAA+2.0 mg•L-1 GA3, 诱导率可达81.27%; 适合腋芽增殖的培养基为WPM+3.0 mg•L-1 6-BA+0.1 mg•L-1 IBA, 增殖系数可达4.26; 最佳生根培养基为1/2MS+1.0 mg•L-1 IBA+0.5 mg•L-1 NAA+1.0 mg•L-1 GA3, 生根率为82.18%。炼苗移栽到泥炭土:珍珠岩:蛭石=2:1:1的基质中, 成活率达76.0%以上, 该结果为木通的快繁提供了一条新途径。

Tissue Culture and Rapid Propagation of Akebia trifoliate var. australis

Abstract:

In order to establish the tissue culture and rapid propagation system of Akebia trifoliate var. australis, the effects of plant growth regulators on plant regeneration in vitro were studied using stems with axillary buds as explants. The results showed that the better disinfection method for stems with axillary buds was to use 75% alcohol immersion 30 s, then use 2% NaClO immersion 15 min; the fittest medium for germination of stem axillary bud was WPM+1.0 mg•L-1 6-BA+0.5 mg•L-1 IAA+2.0 mg•L-1 GA3, with a 81.27% germination rate of axillary bud; the best medium for shoot multiplication was WPM+3.0 mg•L-1 6-BA+0.1 mg•L-1 IBA, the multiplication coefficient could reach 4.26; the best rooting medium was 1/2MS+1.0 mg•L-1 IBA+0.5 mg•L-1 NAA+1.0 mg•L-1 GA3, with a 82.18% rooting rate. After the culture-bottle seedlings were transplanted into the matrix composed of peat soil, perlite and vermiculite (2:1:1), the survival rate was up to 76.0%. The research supplied a new way for rapid propagation of Akebia trifoliate.
